چکیده

Filariasis is a serious health hazard affecting millions of people in tropical and subtropical regions of developing countries including India. The specific diagnosis and control measures are essential for effective management of the disease. It has been shown earlier that the excretorysecretory products of filarial parasitescontain diagnostically important embryo antigens; therefore, it would be important to identify and characterize the embryo stage antigens having potential for serodiagnosis of human filariasis. In the present study efforts were made to analyse the protein and antigenic pattern of embryo stage of Setaria cervi, the bovine filarial parasite. The S. cervi embryos (ScEmb) were isolated by dissecting adult female worms and purified using Percoll gradient. The SDSPolyacrylamide gel electrophoresis revealed15-18 protein bands in S. cervi embryo somatic extract in molecular weight range of 6.5 – 200 kDa. The polyclonal antibodies produced against S. cervi embryo showed significantly high reactivity with ScEmbantigens as well aswith antigens fromadult and microfilarialstages. The 10-14 protein bandsof S. cervi embryo were recognized by anti-ScEmb polyclonal antibodies in immunoblotting. These studies revealed some qualitative and quantitative differences among the antigenic pattern of three stage of S.cervi.The S.cervi embryos exhibited high reactivity with antibodies present in filarial patients’ serathereby suggesting the presence of some antigens common/cross-reactive with human filarial parasite. The protein bands of 60 kDa, 45 kDa, 29 kDa and 27 kDa of S. cervi embryo stage were recognized by filarial patients serum pool and may have diagnostic utility in human lymphatic filariasis Manuscript received January 10, 2016; revised June 2

Evaluation of In Vitro Antifilarial Potential in Alcoholic Extract of Bark of Albizzia Lebbeck

The effect in alcoholic extract of Bark of Albizzia lebbeck was studied for antifilarial activity using Setaria cervi (S. cervi) worm procured from the freshly slaughtered buffalo. We used the whole worm (w.w.) preparation and nerve muscle (n.m.) complex of Setaria cervi (S. cervi). The n.m. complex is made by stripping the cuticle of the worm. The effect was stimulant in nature in both the prep...

Somatic antigens from male and female adults of Setaria cervi: immune reactions in guinea pigs.

Antigens obtained from adult female Setaria cervi were more protective than those of males against the development of microfilaremia in guinea pigs (transplanted with 10 S. cervi). In high dose levels (2 ml 500 microliters/ml) significant resistance was induced by antigens from male (52% of protection) and female (62% of protection) worms. The antigens from female worms were effective (40% prot...

Partial purification and characterization of acetylcholinesterase isozymes from adult bovine filarial parasite Setaria cervi.

Filariasis is a major health problem, affecting millions of people in tropical and sub-tropical regions of the world. The isolation and characterization of parasite-specific enzyme targets is essential for developing effective control measures against filariasis. Acetylcholinesterase (AchE, E.C. 3.1.1.7), an important enzyme of neuromuscular transmission is found in a number of helminths includ...

Secondary antifilarial screening in dogs using Setaria cervi as test organism.

It has been reported (1, 2) that intraperitoneal implantation with two male and two female adult Setaria cervi produces microfilaremia in rats within two weeks and lasts from 54±6 days. When the number of worms implanted is increased, microfilaria appear earlier but the mortality of the host is higher. Complete disappearance of microfilaria from peripheral circulation results after oral and int...

Potential antifilarial activity of fruit extracts of Ficus racemosa Linn. against Setaria cervi in vitro.

Effect of alcoholic and aqueous extracts of the fruits of F. racemosa Linn., on the spontaneous movements of both the whole worm and nerve muscle preparation of Setaria cervi and on the survival of microfilariae in vitro was studied. Alcoholic as well as aqueous extracts caused inhibition of spontaneous motility of whole worm and nerve muscle preparation of Setaria cervi characterized by increa...
